Best Day of My Life ist ein Song aus der vierzehnten Episode der fünften Staffel, New New York, und wird von Blaine und Sam gesungen. Nachdem sich Sam, seit seiner Ankunft in New York, zu einem Coachpotatoe entwickelt hat, macht Blaine sich Sorgen um ihn und spricht ihn, auch auf Anraten Kurts, darauf an. Sam erklärt, dass er keinen einzigen Modelauftrag bekommen hat und meint, dass er New York hasst. Blaine nimmt das zum Anlass, ihn mit nach draußen zu nehmen und ihn vom Gegenteil zu überzeugen. Die beiden laufen den Time Square entlang und performen den Song, wobei Sam, entgegen seiner vorherigen Einstellung, sichtlich Spaß hat.

Das Original stammt von American Authors aus ihrem ersten Album "Oh, What a Life" aus dem Jahr 2013.
