Defying Gravity from Wicked is featured in 100, the twelfth episode of Season Five. It is sung by Kurt, Mercedes, and Rachel. They re-do the song previously sung in Wheels, the ninth episode of Season One, as part of the show's special hundredth episode.
Rachel and Mercedes had been rivalizing ever since their return to McKinley High. While being interviewed on Fondue For Two, Rachel and Mercedes realize that they plan to sing the same song to Glee Club. Although Rachel objects that herself and Kurt wanted to sing the song, Mercedes manages to persuade her to let the three of them sing it, and to make it a diva-off between the two of them.
As a result, Mercedes, Kurt, and Rachel end up singing Defying Gravity in front of the current and past New Directions. They want them to vote for who should win the diva-off, but Will gets them to postpone that until the following day.

- Chris expressed interest in re-doing this song when asked about what song he wanted to do in the 100th episode. Source
- After the performance, April states that "that song should be on the Broadway." This is an ironic reference to portrayer Kristin Chenoweth's long-time role as Glinda in the Broadway production of Wicked, which Defying Gravity is from.
- Rachel explains to Mercedes that she can't sing Defying Gravity for the assignment, because she and Kurt already planned to re-do their duet, remixing it by switching parts so that she could hit the high note at the end. However, Rachel and Kurt only sung the song together in the released versions; in-universe, the two took turns to sing the song.
