Defying Gravity from Wicked is featured in 100, the twelfth episode of Season Five. It is sung by Kurt, Mercedes and Rachel. They re-did the song previously sung in Wheels, the ninth episode of Season One, as part of the show's special hundredth episode.

- Chris expressed interest in re-doing this song when asked about what song he wanted to do in the 100th episode. Source
- After the performance, April states that "that song should be on the Broadway." This is an ironic reference to portrayer Kristin Chenoweth's long-time role as Glinda in the Broadway production of Wicked, which Defying Gravity is from.
- Rachel explains to Mercedes that she can't sing "Defying Gravity" for the assignment, because she and Kurt already planned to redo their duet, remixing it by switching parts so that she could hit the high note at the end. However, Rachel and Kurt only sung the song together in the released versions; in-universe, the two took turns to sing the song.
